The GLP-1 System
Glucagon-like peptide-1 (GLP-1) is an incretin hormone secreted by L-cells in the small intestine in response to nutrient ingestion. It acts on GLP-1 receptors (GLP-1R) expressed in the pancreas, brain, heart, kidney, and gastrointestinal tract. The primary physiological roles studied include glucose-dependent insulin secretion, glucagon suppression, and gastric emptying delay.
Receptor Binding Mechanisms
GLP-1 receptor agonists bind to the GLP-1R, a class B G protein-coupled receptor (GPCR). Binding activates adenylyl cyclase, increasing intracellular cAMP levels. This cascade has been extensively characterised in pancreatic beta-cell models and is the basis for the glucose-lowering effects observed in metabolic research.
