How to Reconstitute Research Peptides

A complete step-by-step protocol for properly reconstituting lyophilized research peptides using bacteriostatic water. Covers supplies, concentration calculations, technique, storage, and safety.

Step-by-Step Reconstitution Protocol

Prepare Your Work Area

Wipe your work surface with a 70% isopropyl alcohol swab and allow to dry. Wash hands thoroughly. Gather all supplies before beginning.

Calculate Your Volume

Determine the volume of BAC Water needed based on your desired concentration. Refer to the table above. Write this down before proceeding.

Sanitise Both Vial Stoppers

Wipe the rubber stopper of both the peptide vial and the BAC Water vial with separate alcohol swabs. Allow to air dry for 30 seconds — do not blow on them.

Draw the BAC Water

Using a sterile laboratory transfer syringe, carefully draw the calculated volume of bacteriostatic water from the BAC Water vial. Remove any air bubbles by gently tapping the syringe and expelling air.

Transfer Slowly Down the Vial Wall

Insert the transfer syringe through the peptide vial septum at an angle. Slowly dispense the BAC Water down the inner glass wall of the vial — never directly onto the lyophilized powder cake. This prevents denaturation from mechanical force.

Dissolve Gently — Do Not Shake

Gently swirl the vial in slow, circular motions until the powder is completely dissolved. This may take 30-60 seconds. Do not shake, vortex, or agitate vigorously — this can cause aggregation and denaturation.

Inspect the Solution

The reconstituted solution should be clear and colourless (or very slightly yellow for some peptides). Discard immediately if the solution is cloudy, contains particulates, or has an unusual colour.

Label and Store

Label the vial with the peptide name, concentration, reconstitution date, and your initials. Store at 2-8°C (refrigerator). Do not freeze reconstituted peptide. Use within 28-30 days.

Frequently Asked Questions

Can I use sterile water instead of BAC Water?+
Sterile water (without preservative) can be used but the reconstituted solution must be used within 24 hours and cannot be stored. BAC Water is strongly preferred for research protocols requiring multiple uses over days or weeks.
What if the peptide doesn't fully dissolve?+
Allow more time and continue gentle swirling. Some peptides (particularly longer sequences) may take several minutes to fully dissolve. If the powder still does not dissolve after 5 minutes of gentle swirling, try adding a small additional volume of BAC Water. Never heat the vial.
Can I re-freeze reconstituted peptide?+
Repeated freeze-thaw cycles significantly degrade peptide integrity. Reconstituted peptide should be stored at 4°C and not refrozen. If you need to store for longer periods, reconstitute only what is needed and keep the remainder lyophilized at -20°C.
How do I know if my reconstituted peptide has degraded?+
Signs of degradation include: cloudiness or turbidity, visible particulates, unusual colour, or an off smell. If any of these are present, discard the solution. Always use within the recommended 28-30 day window.
